概 要:
Founded in 1997, Deltagen, Inc. has developed nearly 1,000 distinct knockout (KO) mouse lines in which individual murine genes have been targeted and selectively deleted, including about half of the known G-Protein Coupled Receptors (GPCR’s). In addition, Deltagen has conducted extensive phenotypic analysis on nearly 750 of these KO mouse lines, including metrics, fertility, hematology and serum chemistry, and behavioral and neurological testing. The gene targets were selected from gene families known to be important in the pharmaceutical industry and amenable to small molecule chemistry and antibody development. Over the past decade, Deltagen’s KO mouse lines have been validated by nearly all of the major pharmaceutical companies in the world and continue to be licensed by pharmaceutical and biotechnology companies, as well as universities and academic research organizations, both for basic research studies and for critical decision-making in target validation and pre-clinical drug discovery efforts. The gene knockout process will be outlined and a description of the phenotypic analysis programs will be provided, including Deltagen’s advanced phenotypic programs in the areas of metabolic diseases (obesity and diabetes) and immunological/inflammatory diseases.
XenoPharm, Inc. has exclusive rights to technologies developed at the University of Sydney and Baylor College of Medicine relating to transgenic mouse lines useful in the study of metabolism of xenobiotics (i.e., foreign substances), including “humanized” mouse models for Cytochrome 450-CYP3A4 and Constitutive Androstane Receptor (CAR), two entities heavily expressed in human livers. These humanized mouse lines have had the murine form of these genes deleted and replaced with the human homologues of these genes. Together, CAR and CYP3A4 are involved in the metabolism of over 50% of xenobiotics by the human liver. The humanized mice are important research tools for analyzing drug toxicity and drug-drug interactions and for predicting their metabolic behavior in humans. Humanized mouse lines are predicted to become critical to pre-clinical drug development, particularly for ADMET analysis. These humanized mouse lines will be described and illustrated in greater detail.
