タイトル;An easy and high efficient technique for mouse genome manipulation by electroporation.

要旨;
Recent advance in genome editing technology by CRISPR/Cas9 system facilitates generation of genome-engineered animals. Especially, the technique for delivery of RNA into the mouse zygotes by electroporation enables researchers to establish easily gene knockout animals without microinjection. However, stimulation by electroporation with standard condition that have been published gave deleterious effects to zygotes and the survival rate of them became much lower than 50% compare to that without any manipulations. To make the survival rate higher without decline of the efficiency of genome editing, we tested three kinds of electroporators and two electroporation chambers, and optimized the condition. In this report, we will present the best combination of devices and the optimal condition for the CRISPR/Cas9 system by electroporation in our laboratory, and in addition, the application of this technique to Cre/loxP and Flp/FRT systems will be shown.
