タイトル;Effect of cholesterol loss from cell membrane on the fertilizing ability of cold-stored mouse sperm.

The cold storage of cauda epididymis, which is a male reproductive organ storing matured sperm, is an efficient means to transport genetically engineered mice alternative to the shipment of live animals. The fertilizing ability of cold-stored sperm decreased in the time-dependent manner. Previously, we reported methyl-beta-cyclodextrin (MBCD) recovered the fertilizing ability of cold-stored sperm. However, the effect of MBCD to activate the sperm is not clear. In this study, we focused on cholesterol loss of cold-stored sperm by MBCD and examined fertilizing ability of the sperm. As a result, MBCD greatly increased fertilization rates compared with control or BSA (control at 0 mM: 19.5%, BSA: 22.5%, MBCD: 79.1%). MBCD indicated the highest percentage of acrosome reacted sperm (control at 0 mM: 36.7%, BSA: 55.6%, MBCD: 68.5%). In addition, MBCD strongly facilitated cholesterol efflux from the plasma membrane of cold-stored sperm (control at 0 mM: 93.3%, BSA: 85.4%, MBCD: 46.7%). These results suggest that the application of MBCD with the IVF improve d fertilization rate by inducing acrosome reaction and promoting cholesterol efflux. We strongly believe that MBCD is useful to improve the fertilizing capacity of cold-stored and transported sperm of genetically engineered mice.
