タイトル;Role of ANGPTL2 in colon cancer cell survival.

Abstract;
Recently, we reported that Angiopoietin-like protein 2 (ANGPTL2) plays an important role in inflammatory carcinogenesis, tumor cell invasion and metastasis (Cancer Res. 2011, 2012, Sci. signal. 2014). However, it has been obscure whether ANGPTL2 affects the tumor cell survival. In this study, we investigated the role of ANGPTL2 on anti-neoplastic drug-induced apoptosis in human colon cancer cell lines. We demonstrated that expression of ANGPTL2 reduces drug-induced apoptosis in colon cancer cell lines and upregulates pro-survival BCL-2 family genes, play a major role in the protection against drug-induced apoptosis. RNA sequencing showed that Syk is induced in ANGPTL2 expressing SW480 cells. Furthermore, we showed that enhancing cell survival activity and induction of BCL-2 family genes by ANGPTL2 depend on Syk-PI3K and NF-κB pathway. These results suggested that ANGPTL2 protects against drug-induced apoptosis in colon cancer cells by regulating BCL-2 family genes through Syk-PI3K pathway.
