Abstract;
Obesity and its-associated metabolic dysfunction predispose individuals to coronary heart disease (CHD); however, mechanisms linking these conditions have not been clarified. Recently, we found that angiopoietin-like protein 2 (Angptl2) is an adipose tissue-derived inflammatory factor causing metabolic dysfunction in obesity.
Here we show that Angptl2 is abundantly expressed in endothelial cells from patients with CHD and its metabolic risk
factors and from mice fed a high-fat diet, whereas Angptl2 is minimally expressed in control subjects without CHD and metabolic
disease. Endothelial cell-derived Angptl2 promoted vascular inflammation through activating the integrin α5β1/NF-κB cascade, promoting endothelial dysfunction and atherosclerosis in mice. Circulating Angptl2 concentrations were positively associated with Framingham risk scores predictive of CHD and the frequency of future primary CHD events based on
a prospective cohort study.
Thus, Angptl2 is an inflammatory mediator linking obesity with CHD, suggesting it could function as a target in novel prevention and treatment strategies for CHD.
