タイトル;Role of the histone demethylase LSD1 in the regulation of hepatic energy metabolism.

Abstract;
Excessive caloric intake lifestyle leads to a failure of energy homeostasis, such as metabolic syndrome. The unbalanced diet causes a change in gene expression levels of enzymes such as lipid and carbohydrate metabolism. And, as a mechanism linking changes in gene expression profiles with environment factors such as lifestyle, we assume that epigenetic mechanisms are involved. However, still very little is understood about the molecular mechanisms.
So, we hypothesized that epigenetic mechanisms are involved in energy environmental adaptation in the liver which plays a central role in energy homeostasis in vivo. By the comprehensive gene expression analysis using microarrays, we found that the expression of genes involved in fatty acid β-oxidation in mitochondria is significantly increased in lysine specific demethylase 1 (LSD1) knockdown cells. These genes were directly downregulated through epigenetic mechanisms by LSD1. Based on these data, we also confirmed changes in mitochondrial activity in LSD1 inhibited cells by JC-1 staining and extracellular flux analyzer.
These results suggest that LSD1 is involved in energy homeostasis through epigenetic mechanisms.
