Abstract;
Embryonic stem (ES) cells are derived from the inner cell mass, which are pluripotent cells and can be maintained in an undifferentiated state, and stimulated to differentiate into various cell types. We have previously described a procedure for inducing embryonic stem cells into regional specific gut endoderm by culturing the ES cells directly on a monolayer of M15, a mesonephros-derived supporting cell line.   
In this study, we established efficient procedure for intestinal epithelial (IE) like cells from ES cells using M15 and MEF. First, we performed definitive endoderm (DE) differentiation of mouse ES cells using M15 cells. These DE cells were isolated by flow cytometry. For intestinal differentiation, sorted DE cells were cultured on feeder cells with two chemicals for several days. Then, Cdx2-positive IE like cells were appeared. These differentiated cells expressed markers of the intestinal differentiated cell types, such as the absorptive enterocyte, goblet cell, enteroendocrine cell and Paneth cell. These cells are functional intestinal cells, as they showed alkaline phosphatase activity. In addition, this procedure is applicable to human ES cells.
In conclusion, using the feeder system, ES cells can efficiently differentiate into not only the pancreatic and hepatic lineages but also intestinal lineage. This procedure is available for investigation of intestinal differentiation and regeneration medicine for intestinal disease.
