Abstract;
In vitro fertilization is a well established technology for mice and is frequently used for reproducing and expanding a colony of genetically engineered mice. It is well-known that the fertilization rates of inbred or hybrid strains of mice are quite different and tend to be strain-dependent. However, the cause of the strain-dependent difference in fertility in mice is still not fully understood. Recently, proteomic analysis is frequently available for discovering a candidate protein playing an important role in various biological events. In this study, we focused on the level of sperm proteins in different strains of mice and identified the sperm protein using a MALDI-TOF-TOF tandem mass spectrometer combined with isobaric tags for relative and absolute quantification (iTRAQ) reagent. We then compared the abundance level of these identified proteins with the representative inbred strains of C57BL/6 mice, DBA/2 mice and their F1 strain of B6D2F1 mouse. As a result, we succeeded in identifying 2488 proteins of sperm that commonly exist in all strains of mice. Quantitative analysis revealed that several proteins are distinctly different between the three strains. The number of 258 and 60 proteins is higher in sperm taken from DBA/2 and B6D2F1 mice than in C57BL/6 mice. On the other hand, the number of 175 and 78 proteins is lower in DBA/2 and B6D2F1 mice than in C57BL/6 mice. In summary, we established a proteomic analysis system using iTRAQ for mouse sperm which revealed a great number of mouse sperm proteins and strain-dependent differences of several sperm proteins between three strains of mice.
