タイトル;Proteomic analysis of phosphorylated proteins related to the chemosensitivity in glioma using 2D-DIGEcombined with phospho-specific staining.

Abstract;
Alterations of chromosome arms 1p and 19q in anaplastic oligodendroglioma(AOG)/oligodendroastrocytoma(AOA) are associated with chemotherapeutic response and overall survival of patients. To clarify the mechanism of the chemotherapy sensitivity of AOG/AOA, we analyzed tissue specific proteins which show the different expressions and phosphorylations in each tumor. The tumors collected after the surgical operation from the AOG/AOA patients, were analyzed pathological and genomic background, and divided into two groups according to their chemotherapy sensitivities. The proteins prepared from each tumor were subjected to 2-D DIGE, combined with ProQ diamond staining. About 4000 spots in total and 1700 phosphorylated proteins were detected in each gel. Ninety-two spots showed statistically significant differences (p<0.05) in two groups. Among these spots, we identified more than 15 spots showing statistically significant differences in the phosphorylation. These proteins include not only proteins reported as glioma markers such as GFAP and HSP90, but also unknown proteins such as NF-L, Rab-GDI, etc. The identified proteins may have a potentiality to be new markers for glioma and specific functions to show sensitivity differences against chemotherapy in the AOG/AOA.
