Abstract;
Myeloid elf-1-like factor (MEF) or Elf4 is an ETS protein, which has a wide distribution in hematopoietic and non-hematopoietic tissues. MEF is known to up-regulate the basal expression of the anti-microbial peptides, lysozyme and human beta defensin-2 in epithelial cells, bind to granulocyte macrophage colony-stimulating factor and interleukin-3 promoters in hematopoietic cells and activate the transcription of perforin in natural killer cells. The numerous target genes of MEF and its biological functions signify the importance of this Ets transcription factor. Here we show that MEF is modified by conjugation with SUMO-1 (small ubiquitin-related modifier) both in vitro and in vivo at lysine 657. This modification led to a decrease in MEF activity on its target gene, lysozyme. Chromatin immunoprecipitation analysis revealed that SUMO-conjugation diminished the recruitment of MEF to the lysozyme promoter, which partly explains the down-regulation of MEF activity by SUMO. Another important modification that commonly occurs in transcription factors is acetylation. We demonstrate here that MEF is also regulated by acetylation and the functional consequence of this modification is the enhancement of MEF activity. Upon treatment with TSA, an inhibitor of histone deacetylase complex, MEF protein and endogenous lysozyme mRNA expressions were increased. These findings provide important information on the modifications that occur on MEF and how these modifications affect its transcriptional activity.
